At NA 1.50, the evanescent field penetration depth in TIRF is minimized to its theoretical limit, providing the tightest possible optical sectioning at the coverslip surface for single-molecule detection, membrane dynamics, and receptor-ligand interaction studies where background fluorescence from out-of-focus molecules must be completely eliminated. The plan apochromat correction ensures flat-field imaging with accurate chromatic registration across all fluorescence channels — essential for multi-color TIRF co-localization, PALM, STORM, and dSTORM super-resolution workflows where chromatic shift between channels would compromise localization accuracy.
