The Olympus LU PLANAPO 25× Silicone is a specialized intermediate-magnification objective that addresses the need for wide-field, high-quality imaging of large biological structures at depth. The 25× magnification with FN 26.5 provides a wide imaging field ideal for organoids, brain slices, embryos, and other large specimens where both field coverage and optical performance at depth are required. The 2.0mm working distance is exceptional for a silicone immersion objective at NA 0.85 — providing ample clearance for thick specimens, imaging chambers, and experimental manipulations beneath the objective. Silicone immersion (n=1.40) closely matches the refractive index of biological cytoplasm and tissue (~1.38), reducing spherical aberration when imaging deep into living specimens and maintaining image quality at depths where water immersion objectives suffer increasing aberration. The super apochromat (PLANAPO) correction ensures accurate chromatic registration across all fluorescence channels, essential for multi-color co-localization and any imaging workflow where chromatic shift between channels would compromise data integrity.
