Fura-2 requires excitation at 340nm and 380nm, with the ratio of emission intensities used to calculate intracellular calcium concentration. Standard oil immersion objectives have significantly reduced transmission below 380nm, making them unsuitable for accurate fura-2 ratiometric imaging. The HC PL APO UV designation indicates that this objective maintains high transmission at 340nm, ensuring sufficient UV excitation intensity for accurate ratiometric calcium measurements.
