DIC microscopy uses a Nomarski prism to split and recombine polarized light beams traveling through slightly different lateral positions in the specimen — converting refractive index and thickness gradients into amplitude contrast to produce a high-contrast pseudo-three-dimensional relief image of transparent, unstained living specimens. The U-DICTH features a rotatable prism that allows DIC contrast to be fine-tuned by rotating the prism within the slider, providing precise control over the bias retardation and the resulting image contrast. The slider offers two mounting options: it can be installed in the nosepiece slot for direct integration with the objective turret, or in the U-KPA slot for alternative positioning within the optical path. As with all Olympus DIC components, the U-DICTH can be installed on any brightfield transmitted light microscope capable of accepting polarizing filters and the appropriate condenser and objective prisms.
